Clone a gene

Copy a gene, put it into a plasmid, get it into bacteria and confirm the sequence: the classic cloning workflow.

0 of 8 steps practisedStart with step 1: PCR →
  1. PCRNot started

Amplify the gene from template DNA with primers that add the ends you need for cloning.

  2. Check that the PCR gave one band of the right size before you go further.

  3. Cut the insert and the vector so their ends match.

  4. Join the insert into the vector at the right insert to vector ratio.

  5. Get the plasmid into competent E. coli and select the cells that took it up.

  6. Grow a positive colony and purify its plasmid.

  7. Measure how much plasmid you have and how pure it is before sequencing.

  8. Read the insert base by base to confirm there are no mutations.
